I. Historical Perspective & Models (Sections 6.1 - 6.2)
The Meselson-Stahl Experiment: Proved the
Semiconservative nature of replication, where each daughter
DNA molecule consists of one parental strand and one newly
synthesized strand.
Visualization of Replication: Early studies using bacterial
models demonstrated that replication starts at a specific Origin
of Replication and proceeds bidirectionally.
Fundamental Rules:
Synthesis always occurs in the 5' to 3' direction.
The process requires a template strand and a free 3'-OH
group to add nucleotides.
