All Cyclooxygenase Inhibitors Acetaminophen - not anti inflammatory Ace- enzyme Inhibition of the prostagladin process In general, the effects of prostaglandin -are not all negative effects Classification COX inhibitors 1st and 2nd gen- based on Selectivity 1st gen- Inhibits all COX 2nd gen- target the bad prostaglandin One is considered more helpful and one is more related to inflammation BB 1st and 2nd gen- Selectivity made the difference COX inhibitors and acetaminophen • COX 1 “good” – Housekeeper • Cox 2 “bad” – Works at sites of inflammation and pain – Brain :fever and pain – Kidney: supports renal function – Blood vessels: vasodilator – Colon: contributes to colon cancer Some enzymatic action is a good thing- stomach, kidneys (good and bad) and platelets (stim. platelet aggregation)
