Chromatography is a process for separa ng components in a mixture and may also be used
to iden fy the components in a mixture. All types of chromatography have a both a
sta onary phase with them being solids such as silica, paper, or alumina. A mobile phase
which is a liquid, such as water, there are mul ple types of chromatography that are used
within the laboratory for a wide range of reasons, such as thin layer chromatography, gas
chromatography and paper chromatography etc.
Paper chromatography is commonly used in the laboratory as an analy cal technique used
to separate mixtures of soluble substances, such as ink or dye. There are two stages to paper
chromatography the rst being the sta onary phase, the sta onary phase consists of when a
solid such as glass, silica or paper and remains xed in place and on which various solutes
are absorbed to varying degrees.
Thin layer chromatography is used to separate mixtures of separate soluble substances, this
is the same with paper chromatography as all types of chromatography is used to work on
the same principle of separa ng mixtures. Silica gel is used in the sta onary phase which is
xed in place and the mobile phase being a liquid or a gas.
Aim
The aim of the paper chromatography inves ga on was to iden fy and separate coloured
mixtures, such as water-soluble markers. The aim of the thin layer chromatography
inves ga on was to iden fy compound within a mixture, and also determining their purity.
Hypothesis
If there is a water-soluble solvent present, only the water-soluble pigment would travel
along the chromatography paper.
Risk assessment
Ethanol is ammable due to its chemical proper es; safety precau ons would have to be
taken to prevent a re. If ethanol was to be inhaled it could cause irrita on to the throat and
the nose, to prevent this from occurring the handling of ethanol would be completed in a
fume cupboard. If ethanol was to contact the eyes, it would cause severe irrita on to the
eye if not washed out at a designated sta on. To prevent this from occurring, safety goggles
would have to be worn throughout the dura on of the prac cal. Ethanol is also an irritant to
the skin, gloves would have to be worn throughout the dura on of the prac cal.
Ethyl acetate is also a ammable liquid due to its chemical proper es; so, safety precau ons
would have to be taken to prevent a re. If ethyl acetate was to contact the skin, it could
cause severe irrita on if not washed o with soap and water. To prevent irrita on caused by
skin contact, gloves would have to be worn throughout the dura on of the prac cal. If ethyl
acetate was to be inhaled it could also cause irrita on to the nose and throat, to mi gate
this risk ethyl acetate would have to handled in a fume cupboard to reduce the inhala on
risk of ethyl acetate. As ethyl acetate was to contact the eyes it would also cause irrita on,
to mi gate this risk safety goggles would have to be worn during the prac cal.
