Part 6 tests β-galactosidase enzyme activity across pH values (6.4, 6.8, 7.2, 7.7, 8.0) using ONPG substrate (colorless until cleaved, releasing yellow ONP). Absorbance measured via spectrophotometer at 420 nm: highest absorbance at pH 6.8 (0.193), indicating optimal enzyme activity. pH extremes denature enzymes (unfold) and reduce activity. Conclusions: buffers resist pH changes, the Henderson-Hasselbalch equation predicts buffer pH, and enzymes have optimal pH ranges for function. Suitable for college chemistry and biochemistry students learning buffer systems, pH resistance, enzyme kinetics, and spectrophotometry.
