BIOS 225 Lab Report Week 2- Transferring Microbes While Using Aseptic Technique Introduction: Purpose: To learn an aseptic technique to reduce the minimum contamination, while transferring bacteria from one place to another place by using inoculating loop or a needle, as well as to isolate bacteria colony using the streak method. Materials and Methods: Test tubes, bacteria (water), tops to test tubes, tube rack, LB agar plates, media solution, Bunsen burner, and transfer loops. Safety equipment included goggles and gloves. Aseptic technique: a method designed to prevent contamination from microorganisms. Results and Discussion: During this lab, the instructor explained how to use aseptic techniques while transferring bacteria from one area to another. The instructor also demonstrated how to transfer bacteria from tube to tube as well as from tube to agar plate using aseptic technique. In order to practice aseptic technique and transfer bacteria, the following steps must be followed. Transferring from tube to tube steps using aseptic technique: (a) Turn on Bunsen burner and sterilize loop over flame (until loop turns red). (b) Take top off bacteria, warm opening of bacteria tube, then put loop in tube to get a sample (c) Place top back on to bacteria tube. (d) Remove top from media tube, warm opening of media tube, then place bacteria sample from the loop into media. (e) Place top on media (f) Use Bunsen burner to re-sterilize loop. Transferring bacteria from tube to LB agar plate to conduct streak test using aseptic technique: (a) Turn on Bunsen burner and sterilize loop over flame (until loop turns red). (b) Take top off bacteria, warm opening of bacteria tube, then put loop in tube to get a sample (c) Place top back on to bacteria tube. (d) Take top off LB agar plate and choose one side to do the first streak. (e) Second streak, make sure to send loop through streak one. (f) Third streak, make sure to send loop through streak two but not streak one. (g) Fourth streak, make sure to send loop through streak three but do not touch first or second streak. The fourth
