The objective of this study was the evaluation of the effect of changes in levels of
endothelial cell-derived inflammatory mediators IL-8 & ICAM-1 in response to IL-1βand
IFN-γ and their implications in Multiple Sclerosis (MS). To strengthen my statement
series of tests were conducted. For this, I evaluated changes in the levels of endothelial
cell-derived inflammatory mediatory IL-8 in response to IL- 1β and IFN-γ and their
collective effects on MS. By this study cytokine, treatment was IL-1β and IFN-γ
determined. ELISA was done for the detection of IL-8 level.
1. Enzyme Immuno-Absorbent Assay (ELISA) For The Detection Of IL-8
Levels
Human IL-8 level was checked using an ELISA kit. The method of ELISA was followed
as recommended. After performing the whole procedure, the absorbance was
measured at 450nm. Values of positive and negative control were also measured at
450nm matched with the mentioned values on the literature of the ELISA kit for IL-8.
Data was collected. Data were analyzed statistically using ANOVA by Minitab. Here
results were represented graphically.

This graph interprets the values of IL-8 concentration in pg/ml with their absorbance at
450nm. As the graph is an almost straight line which shows that as the dilution of IL-8
increases the value of absorbance increases. The concentration of IL-8 increases when
, both treatments are used together. As the value of R 2 falls nearly equal to 1 (0.979)
which shows that the experiment conducted predict that the model used is highly
predictive.

This graph shows the relationship between dilution and its effect on the concentration of
IL-8 and INF production by human endothelial cells. It also shows no synergetic effect
as IL-beta and INF+IL-beta values are the same. Control was intact shows ELISA was
performed in ideal conditions. This shows that the value of INF+IL-Beta combined is not
statistically significant at dilution 1:2.
