Introduction: Our skin forms the first line of defense. It has several characteristics that make skin an ideal environment for the growth of most microbes. Some of the characteristics of skin are high percentage of salt, low pH, sebum and an outer layer of dead skin cells that act as a barrier to pathogens. The only organisms that can tolerate high levels of salt are able to grow on skin. Purpose: The purpose of this lab is to learn how to isolate and identify bacteria from skin and learn the characteristics. Also, to learn which selective mediums select for salt tolerant bacteria and reveals mannitol fermentation. Materials: Gloves Apron Goggles Test tube rack MSA plates Nutrient agar plates Sterile swabs Black China marker An Inoculating loop Ruler 1 Bottle of propane 1 Bunsen burner 1 Sparker Pure culture of P. fluorescens Pure culture of E. coli Pure culture of S. marcescens Pure culture of S. epidermidis Methods: Before beginning the experiment put on apron, goggles and gloves for your own protection. o For this experiment we will be working in groups of four o Obtain 2 MSA plates and 2 nutrient agar plates per group o Take one MSA plate and divide it in four parts o Label each quadrant correctly with the name of the bacteria and your groups initials on each plate o 1.
