Question 1: The following analgesics (Caffeine, Paracetamol, Naproxen, and Aspirin) were
separated on 3 C18 50 x 4.6 mm column using H2O with 0.1 %glacial acetic acid/MeOH
(72:28 v/v). Detection was carried out using UV at 275 nm.
A) What will be the expected order of elution of the drugs in the chromatogram (show on
chromatogram)?
B) Justify the order of elution (draw the structure of each drug)

Looking at the structures of the four drugs its obvious hat naproxen has the lowest polarity
as it only includes two possible ionizing groups but two ring system. Acetaminophen has.
OH and an Amid group which is more polar than the carboxylic group of Aspirin. Caffeine
on the other hand has one pie deficient nitrogen and two carbon-oxygen double bond ( pie
deficient makes good ionization).
