1. Which step in H & E staining is for deparaffinnization?
- Deparaffinization is the removal of excess paraffin in the tissue and it is done at the first or initial
xylene bath. The tissue will be immersed in xylene for 3 minutes each in 2 changes.
2. Why are several changes of xylol used at the start of the staining?
- Several changes are required at the start of staining for further deparaffinization. There is a need
to do this in order to ensure the full removal of excess paraffin present in the tissue which will
allow excellent staining of the specimen.
3. Why are several changes of xylol used at the end of staining?
- The final step in H&E staining is last xylene bath which clears the specimen prior to mounting.
This step will ensure the completeness of the process and it will make the specimen ready for
mounting.
