BS02 Basics to Microscopy
Objective: To become familiar with the basic components of a light microscope as well as how
to load a sample for viewing.
Procedure:
1. Review parts and components
2. Load sample slide onto microscope
3. Select magnification
4. Make necessary adjustments to optimize sample visualization
Basic Parts of Microscope:
Eyepiece: one most microscopes you can grab and
pull them apart/together to fit the width of your
eyes. You should only see one single circle. If you
see two circles you may need to either compress
or expand the eyepiece.
Arm/neck: thick curvature of microscope
o If transporting microscope grab by
arm/base
Objectives: lenses that provide part of the
magnification
o Shortest objective: 4X (red) DRY
o 10X (yellow) DRY
o 40X (blue) DRY
o Largest objective: 100X (white) OIL
Revolving nosepiece: has different objectives with different powers of magnification
o By grabbing and rotating you can switch objectives
o Will hear click when objectives are locked in place
Stage: flat surface where you place your sample
Stage controls: clamps that hold sample onto stage
o How to load slide onto microscope:
Pinch clamps together to open the arms on the stage, push the slide on
and slowly release
Stage clips: lift them up and gently set them down on the glass cover slide just to hold it
in place
o Will ensure that sample is not moving around while you’re trying to find the
image
Coarse/fine focus: focus knobs on the side of the microscope
