BS01 Introduction to Medical Microbiology
Objective:
1. Cultivation of samples (growth conditions)
Equipment used
2. Identification of samples (biochemical assays)
Tests available
3. Evaluation of samples (microscopy)
Visualization and recognition of key characteristics
Basic equipment:
1. Cleaning: need to work in sterile environment
Autoclave: 125C
i. Uses heat, pressure and steam to sterilize environment
ii. Steam is more efficient than dry heat to achieve a sterile environment
steam takes a couple of minutes compared to dry heat takes
several hours
iii. make sure that autoclave has been depressurized before opening
2. Growing
Fixed incubator: 37C
i. Holds Petri dishes and liquids cultures
ii. Door airtight sealed, has metal door to keep dark conditions
Shaker incubator: 37C
i. Metal rings, flask used for growth media (liquid nutrient broth) to
inoculate bacteria
ii. Once bacteria is in growth media, the shaker incubator shakes and rotates
the culture in a swirling fashion to aerate it to help bacteria grow

4. Storing
Refrigerator: 4C
i. Storing helps preserve samples and keep them for long term
ii. Ex: Petri dishes, test tubes racks
