what do biologist use to characterize and manipulate DNA molecules? how are sticky ends usefull in DNA fragments? restriction endonuclease;;;;; because DNA fragment from different sources but with the same complementary ends can be pasted together to form new hybrid molecules what is pBR322, that we used in lab? a plasmid which are small circular DNA molecule found in bacteria and yeast we did we do in lab to the pBR322 plasmid? we digested it in 3 different restriction enzymes and then separate the resulting fragments using a technique call agarose gel-electrophoresis how does the gel-electrophoresis catch the DNA fragments? because the gel contains a nucleic acid stain, which binds DNA and is fluorescent what did we do in procedure 1? we made 3 test tube each with 2.0 ul Genomic DNA and 3.0 ul of buffer...... we added 3.0 ul sfi l of enzyme to tube 2 and 3.0 ul Msp 1 enzyme to tube 3. No enzyme in tube 1 what did we do in procedure 2? we made 4-9 tubes that had 2.0 ul pBR322, a cetain amount of water, 3.0 ul buffer, and 3 up of either enzyme A, B, C, or a combination of those. what did we do after the rubes were made? we put the tubes in a microcentrfuge so everythin can go to the bottom, and placed them in 37 C for 30 minutes why is it important to load you gel fast? after the 30 minutes is done, what do we do and how much of the sample do we put into the gel electrophoresis? becasue letting the gel sit for a long time with the power off will give you poor results;;;;;;; dilute iwth 30 ul of water and mix, then add only 20 ul of sample into the gel electrophoresis what is pBR322? its circular and has a total of 4361 base pairs
