, PRACTICAL (1)
Microscopic examination of Blood Film
It is a test used in clinical evaluation of the characteristics of blood. It is
performed by smearing a drop of blood on a slide.
Materials :
1-Blood sample (a drop of blood is enough). 2- Sterile disposable lancets.
3-Alcohol pads. 4- Paper towels. 5- Disinfecting solution.
6- Clean microscope slides. 7- Rack for putting slides on.
8-Distilled water in dropper bottle.9- Wright's stain in dropper bottle.
10- Microscope & oil for the “oil-immersion lens".
Procedure :
1. Clean your hands thoroughly with disinfecting solution.
2. Prepare the palmer surface of your non-dominant hand’s fourth finger
or thumb for puncturing by dabbing it with a fresh alcohol pad.
3. Using a sterile, disposable lancet, puncture the palmar surface of the
finger you chose. Use a quick firm stroke.
4. Place a drop of blood from the punctured finger on a clean
microscopic slide about 2cm away from one end.
5. Hold the end of another clean slide at a 50-degree angle against the
first slide and push the blood drop away from the middle of the first
slide. This maneuver will spread the drop of blood to form a line of
blood where the two slides meet. Now push the second slide toward
the middle of the first slide. This makes an even smear across the first
slide. Stop before reaching the end of the first slide.
Dispose the spreader slide in the dustpan
6. Dry the smear of blood by moving it in the air as quick as possible.
7. Place few drops of Wright’s stain counting the number of drops as you
are putting the stain.
8. Wait 4 minutes and then add equal number of drops of distilled water
to the smear. Wait for10 minutes.
9. Rinse the slide gently under tap water for 30 seconds.
10. Dry the slide gently using a towel paper.
