INTRODUCTION
The fungal cell wall consists of,
MANNOPROTEINS
POLYSACCHARIDES – β ( 1, 3 ) linked glucans
β ( 1, 6 ) linked glucans
CHITIN – it protects the fungus from the hostile
environment
, underneath this, lies Ergosterol
Ergosterol acts to maintain the membrane intergrity in the same capacity as
Cholestrol
Ergosterol – first target site
AMPHOTHERICIN B ( AMPH B )
Amph binds with ergosterol in the cell wall and forms pores
This causes rapid leakage of the intercellular ions ( k+, Ca++, Na+ ) which leads
to, cell death
But, it also makes pores in the renal membrane and epithelial cells which, leads
to, NEPHROTOXICITY
NYSTATIN
It has same mechanism of action as Amph B
It is not absorbed from the mucous membrane and skin, which is the reason of
its limited treatment of superficial candida infections
