Title: Gel Permeation chromatography
Introduction: We conducted an experiment called Gel permeation chromatography; Gel
permeation is used to separate mixtures using a chromatogram.
Aim: The aim of the practical was to identify the presence of glucose and starch.
Hypothesis: I hypothesise that if the substance contains starch when iodine solution is added a
colour change from colourless to a dark blue or black should happen. Whereas if the substance
containing glucose then when Benedict solution (blue) is added then the solution should change
from a blue colour to a light red or orange colour.
Equipment:
Plastic filter funnel
Test tube Rack
Test tubes
Water bath
Test tube holder for the water bath
Column
Starch and glucose mixture
Sodium chloride
iodine solution
Sephadex
Benedict´s reagent
Pipettes
Burette stand
Beaker
Lab coat
Risk assessment:
In this experiment there were risks that were avoided using PPE and being careful. The
PPE used were the gloves to protect the hands from the acids ;the amino acid can turn the
hands so the gloves are essential, and the lab coat had to be worn all the time during the
experiment, to prevent any clothes from getting dirty ,ruined or burned. In the experiment
we used a hot water bath; the hot water bath was placed in one corner of the room for the
safety of the students. We used glassware in our experiment we had to be very careful to
prevent any cuts and blood, for this all the table had to be tidy and clean.
