Objective: to become familiar with the basic components if a light
microscope as well as how to load a sample for viewing
Procedure:
1. Review parts & components
2. Load sample slide onto microscope
3. Select magnification
4. Make necessary adjustments to optimize sample
visualization Notes:
Magnify glass, lens, light source, object to look at Basic parts of
microscope: eyepiece, arm/neck, objective, revolving nosepiece,
stage, stage controls, stage clips, course/fine focus, iris
diaphragm, light source, base.
Standard light microscope using halogen oculars You should see a
single circle in the viewfinder.
The adjustment knob is on the left side. You can roll the wheel on
the diaphragm for adjustment 10X yellow, 40X blue, 4x, 100x.
The Types of objections are dry vs. oil—oil is required for 100x.
One drop of immersion oil Intensity of light source: too bright=
saturation, too dark= low visibility
(power of objection) *(power of eyepiece) =total magnification
