Title: GE03 Mounting Techniques
Objective: Microscopic explanation of bacterial samples through
various staining techniques.
Identify the colour and shape of given samples.
Procedure:
For a dry mount:
1. Clean slide (70%ethanol)
2. Circle area on the slide for easy location of the specimen
(optional)
3. Apply the organism to the slide: If it is from culture, use a
sterile loop to spread it onto the slide. If it is from the plate, use
a sterile loop to pick colonies and mix them with a drop of
distilled water.
4. Air dry at room temperature until all moisture has evaporated.
Wet Mount:
1. Clean slide (70%ethanol)
2. Circle area with a wax/hydrophobic pen
3. Apply the organism to the slide: If it is from culture, spread
it onto the slide using a sterile loop. If it is from the plate, use
a sterile loop to pick colonies and mix them with a drop of
distilled water.
4. Wet mount is ideal for viewing the motility of an organism;
