Root tip squah experiment
Method:
1. Get the broad bean at cut off 5mm of the top of the growing root.
2. Put the root tip into a watch glass that has acetic orcein stain and 1.0mol dm-3 hydrochloric acid,
using the proportion 10 parts of stain to one part of acid.
3. Warm up (not boil) it on a hotplate for five minutes.
4. Get a clean microscope slide and put the stained root tip onto it, then proceed to cut it half
transversely and remove the half that is the further away from the apex.
5. Now get acetic orcein and ad three drops of it to the root tip that is on the slide.
6. Use a needle to break the root tip and spread it out equally over the slide.
7. Add a coverslip on top of the root tip, cover it with filterpaper and then press down softly.
8. Use a hotplate to warm the slide up, warm it for 10 seconds. This will increase the staining.
9. Look for the stages in mitosis.

Chromosones during mitosis
In interphase mitosis the chromatins state is least condensed, it looks loosely distributed thoughout
the nucleus. In prophase the chromatin condensation begins which makes the chromosones visible,
the chromosones are condensed through the various stages of mitosis.
In prophase of mitosis, the chromatic condenses, it becomes increasingly compact and coils. This
results in visible chromosones being formed.
