Nitrogen fixationCorrect! Osmotic pressure pH Hydrostatic pressure Quorum sensing Chapter 6: Microbial Growth and Nutrition Question 10 (TCO 4) During the _____ of growth, new cells are being produced at the same rate as other cells are dying. lag phase log phase death phase stationary phase intermediate phase Chapter 6: Microbial Growth and Nutrition Question 11 (TCO 4) Compare and contrast the streak-plate method of isolation with the pour-plate method of isolation. Your Answer: Streaking technique is used to isolate the bacteria and form pure bacterial cultures, it is the simplest form of bacterial isolation, streaking is generally done by the sterile loop or cotton swab. The three streaking techniques are T-streak technique, Continuous streak and Quadrant streak technique. As for pour-plate method of isolation, the diluted samples are poured into the petri dish, allowed to dry for few seconds and then incubated. This techniques is quantitative, we know the dilution of sample and the number of colonies obtained are counted. The dilutions are continued until a countable number of colonies are obtained. This technique needs repeated inoculation of the sample to isolate the required bacteria. Chapter 6: Microbial Growth and Nutrition The streak-plate and the pour-plate methods of bacterial isolation are both used to produce pure cultures of bacteria from specimens. Both techniques involve the use of agar-based growth media contained in Petri plates. Additionally, both techniques involve the use of dilutions as a means of isolating single cells or groups of cells that then grow into isolated colonies. But there are several significant differences between the two techniques. The main difference is the way in which the specimen is diluted. In the streak-plate method, the specimen is diluted by use of an inoculating loop that spreads organisms over the surface of the agar. Thus, colonies appear only on the agar surface. In the pour-plate method, however, dilutions of the specimen are made in tubes of broth and then each dilution is added to melted agar, which is then poured into Petri dishes. Thus, microbes are mixed throughout the agar, and colonies will appear both in and on the medium. Quiz Score: 28 out of 30
