PURIFICATION OF
POLYSACCHARIDS
, ISOLATION AND EXTRACTION METHODS OF POLYSACCHARIDES
• The principle to select isolation method of polysaccharide is to keep the intrinsic properties of
polysaccharides unchanged during the process of isolation and purification.
• Besides traditional crushing methods, ultrasonic gasflow crushing technology is used to ruptu
the cell wall of animal/plant or fungalin recent years,
• Extraction method of hot water
• polysaccharides have bigger solubility in hot water.
• Polysaccharide is stable in hot water, so polysaccharide can receive the minimal damage using this
extraction method. The usual practice is to extract 2 ~ 6 h using hot water
, • Extraction method of dilute alkali-water solution
• Some acidic polysaccharides, or high MW polysaccharides are not easily dissolved
• in hot water. Their solubility is generally bigger in dilute alkali solution than in hotwater.
Therefore, NaOH solution or NaCO3 solution of 5% ~ 15% (w/w) is often usedto extract inste
of hot water.
• When using dilute alkali solution to extract, extraction temperature should be kept below 10 ℃
Otherwise, polysaccharides are prone to degradation.
